MECHANISM OF ACTION AND LITERATURE

Clearing neurotoxic amyloid beta (Aβ) remains one of the biggest challenges in Alzheimer's research. Mitochondrial damage-mediated oxidative stress has been implicated in the progression of several neurodegenerative diseases, including Alzheimer's disease. Studies have shown that Resveratrol, in various experimental models, can suppress high oxidative stress mediated by free radicals despite SIRT1 by maintaining cellular homeostasis. There is compelling evidence that RSV facilitates the attenuation of the expression of BACE1, the rate-limiting enzyme involved in the production of amyloid plaques, and helps reduce the pathological dominance of amyloid plaques. Consequently, the neuroprotective potential of Resveratrol linked to the SIRT1 pathway is widely considered in treatment protocols for Alzheimer's disease. Among natural compounds, Resveratrol holds a special place in attracting many researchers to work on therapeutic applications in Alzheimer's disease and similar conditions. Studies have shown that Resveratrol can suppress oxidative stress and free radical levels, mimicking the effect of calorie restriction (CR), and increase the expression of genes such as AMPK and SIRT1 to maintain homeostasis in animals. While unmodified Resveratrol appears to have poor bioavailability, several studies have clearly demonstrated the in vivo neuroprotective properties of polyphenols derived from red wine. This strongly supports the idea that natural metabolites of Resveratrol may possess biological activities. Furthermore, recent findings have shed light on Resveratrol's potential role in transcriptional and degradation-dependent antiamyloidogenic mechanisms, suggesting that natural metabolites or potent synthetic analogs of Resveratrol may have therapeutic potential in preventing and/or treating Alzheimer's Disease.
